IPTG induciable lac promoter. The sequence is modified a RiboJ -sequence. The promoter has been proven to have a uniduced activity of 50% of the iduced expression.

Usage and Biology

This promoter can be used to effectively express your gene of interest, and also gain some data on its burden on the cell system by inducing the expression with IPTG. It is essential to know how much the different rates of expression affect the viability of the cells and if the increased transcription correlates to increased translation and increased activity.

The sequence is modified with a RiboJ sequence, which is a self-cleaving ribonuclease element. RiboJ cleaves the part in mRNA between 5' UTR and the starting codon so that the mRNA molecules are always the same size. Cutting the extra sequence also makes the mRNA less stable, which decreases the unspecific background when inducing the activity or when measuring the uninduced signal.
